Despite its place in a peptide catalogue, 5-Amino-1MQ is a small molecule: it blocks nicotinamide N-methyltransferase (NNMT). That enzyme moves a methyl group from S-adenosylmethionine onto nicotinamide, yielding 1-methylnicotinamide — a step that spends a methyl donor and, at the same time, removes nicotinamide that could have been recycled into NAD-plus. NNMT is strongly upregulated in the fat tissue of obese animal models, so inhibiting it is the natural experiment for asking whether this methylation flux actively drives sluggish metabolism or simply accompanies it.
