## Description

ARA-290, or cibinetide, is an 11-residue peptide patterned on the outward-facing surface of helix B in erythropoietin. EPO has two separate activity profiles: the well-known blood-forming one, which runs through the classical receptor homodimer, and a tissue-protective one ascribed to a complex of the EPO receptor with the beta-common receptor — the so-called innate repair receptor. ARA-290 was engineered to activate the latter while leaving the former alone.

This design goal explains why the peptide keeps turning up in studies of neuroprotection and inflammation instead of haematology. Its sequence comes from a surface patch of EPO that plays no part in binding the classical receptor, and published studies report no relevant change in red-cell production — eliminating a significant confounder in tissue-protection work.
