## Description

Enclomiphene is the trans geometric isomer of clomiphene, a selective estrogen receptor modulator of the triphenylethylene family. Clomiphene citrate, as traditionally made, contains two isomers whose pharmacology differs sharply: the cis-isomer zuclomiphene acts mostly as an estrogen receptor agonist and lingers with a long elimination half-life, whereas enclomiphene tends towards antagonism and is cleared much more quickly. Separating out the trans-isomer was intended precisely to pull those two profiles apart.

At the mechanistic level, enclomiphene blocks estrogen receptors in the hypothalamus and pituitary, breaking the negative feedback that estradiol normally applies to gonadotropin-releasing hormone signalling. The effect examined in the literature is a rise in luteinising hormone and follicle-stimulating hormone release and, further downstream, higher endogenous testosterone in male models. That sets it apart from the peptide secretagogues and gonadotropins it is frequently compared with, which work through their own receptors instead of by lifting feedback inhibition.
