Semax is a synthetic seven-residue peptide based on the 4–10 fragment of adrenocorticotropic hormone (ACTH), with Pro-Gly-Pro attached at the C-terminus. That extension is the heart of the design: ACTH(4-10) on its own is broken down in plasma within minutes, while the proline-rich tail markedly slows enzymatic degradation. Importantly, trimming ACTH this way removes the part responsible for its corticotropic effect, so Semax is investigated as a neuroactive rather than a hormonal peptide.
