## Description

SLU-PP-332 is a synthetic small molecule — not a peptide — that switches on all three estrogen-related receptors (ERRα, ERRβ and ERRγ). These orphan nuclear receptors control the gene programme behind mitochondrial biogenesis, oxidative phosphorylation and fatty-acid oxidation in muscle. Their name is misleading: they neither are estrogen receptors nor bind estradiol, having been named for a resemblance in sequence, and they work mainly together with PGC-1-family coactivators.

Interest in the compound arose because ERR activation turns on many genes that endurance training also induces, which is why papers have called it an “exercise mimetic”. Researchers add an important qualification: all published data come from cell systems and mice, with no human evidence at all. That gap is precisely what makes it a good probe — it lets labs work out which parts of the exercise-induced transcriptional response depend on ERR signalling and which do not.
