NMN

NMN is the direct NAD+ precursor formed by NAMPT and turned into NAD+ by NMNAT enzymes. Its chemistry, the transport debate and the research context.

NMN (nicotinamide mononucleotide) is the direct biosynthetic forerunner of NAD+ in the mammalian salvage pathway. NAMPT combines nicotinamide with PRPP to form NMN, which NMNAT enzymes then adenylylate to NAD+ in a single additional step. NAMPT is the slowest, rate-limiting enzyme of the route, so providing NMN beyond that bottleneck is the rationale for using it in metabolic experiments.

NMN in the research literature

Rodent studies published from roughly 2011 reported higher NAD+ in tissues together with changes in insulin sensitivity and in mitochondrial and vascular measures after NMN was given; a handful of human trials since 2020 describe biomarker shifts alongside modest, inconsistent functional results. One mechanistic question remains open — transport. NMN is phosphorylated and polar, and some evidence suggests it is dephosphorylated to nicotinamide riboside at the cell surface before entering, while other studies point to a dedicated transporter (Slc12a8). Because of that disagreement, NMN, NR and NAD+ itself are often tested side by side instead of being treated as equivalent.

Regulatory note: since 2022 the US classification of NMN as a dietary-supplement ingredient has been in dispute, which is one of the reasons it is offered here purely as research-grade material.
