GHRP-6 was developed in the 1980s from met-enkephalin analogues, long before ghrelin had been found, and work on it led scientists to the receptor now called the growth hormone secretagogue receptor. Ghrelin was identified later as that receptor's natural ligand.

GHRP-6 lacks selectivity — and that is exactly why researchers still use it. Besides releasing growth hormone, it strongly stimulates appetite via central ghrelin receptors and, at higher exposure, measurably raises cortisol and prolactin. Groups investigating feeding behaviour, orexigenic pathways or the wider pharmacology of this receptor often choose it over the cleaner second-generation peptides for that very reason.
