Sermorelin — GRF(1-29) amide — consists of the first 29 amino acids of human growth hormone-releasing hormone, the shortest stretch that keeps full activity at the receptor. Unlike the engineered analogues elsewhere in this category, it has no substitutions and no acyl chain: the sequence is the natural one. That makes it the nearest available stand-in for the body's own GHRH, and it explains why pituitary-function research still treats it as the reference compound whenever an unmodified ligand is preferable to a stabilised one.

This pentapeptide agonist of the growth hormone secretagogue receptor was developed at Novo Nordisk and found to be remarkably selective: in its original pharmacology it released growth hormone while barely touching ACTH, cortisol or prolactin, in contrast to the older GHRP-6 and GHRP-2. What separates sermorelin from CJC-1295 (No DAC)?
Both are 29-residue GHRH fragments, yet sermorelin keeps the natural sequence whereas CJC-1295 (No DAC) has four DPP-4-resistant substitutions. Sermorelin therefore acts more briefly and more closely resembles endogenous GHRH — why it is still the reference ligand in pituitary research.
What does pairing a GHRH fragment with ipamorelin achieve?
The two hit different receptors on one pituitary cell: sermorelin the GHRH receptor, ipamorelin the ghrelin receptor, which also dampens somatostatin tone that would otherwise limit the response. In animal and cell models, the combination produces a bigger measured growth hormone pulse than either peptide alone.
Is each peptide independently verified?
