Cells make myostatin as a longer precursor, which is then cut in two: the mature growth factor — a C-terminal dimer — and an N-terminal prodomain called the propeptide. Rather than simply falling away, the propeptide stays attached to the mature dimer, holding it in a latent complex that cannot activate its receptors until BMP-1/tolloid proteases release it. Adding the propeptide on its own is therefore the most natural way to switch off myostatin signalling in an experimental model — which is why it is used as a point of comparison for engineered ligand traps like ACE-031 and for follistatin.
The signalling route it interrupts is well understood. The mature ligand — two 109-residue chains linked by a disulfide — binds the type II receptors ActRIIB and ActRIIA, recruits ALK4 and ALK5 as type I receptors and signals by phosphorylating Smad2/3. As the propeptide acts on the ligand and not on the receptor, other ligands of ActRIIB are unaffected — a difference in selectivity that makes it a useful parallel arm next to ACE-031, which captures a far broader range. Each vial holds 1 mg of freeze-dried recombinant protein with purity of 95% or more by SDS-PAGE and HPLC; dissolve it gently in sterile or bacteriostatic water, add a carrier protein to dilute stocks and portion it out before freezing instead of thawing it repeatedly.

How does GDF-8 differ from the myostatin propeptide?
GDF-8 is myostatin itself — the active dimer that does the signalling. The propeptide is the N-terminal piece cut from the same precursor; it remains bound to the active dimer and keeps it switched off. Researchers therefore use it as the ligand's built-in inhibitor, not as the ligand.
Where does it sit relative to ACE-031 and Follistatin-344?
All three shut down the same pathway, each at its own point. ACE-031 is a lab-built, soluble version of the ActRIIB receptor that soaks up many ligands; follistatin is a naturally occurring protein that binds activins and myostatin; the propeptide is the part of myostatin that normally keeps it latent. Because their selectivity differs, studies often run them as parallel arms.
